This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The major goal of this project is to improve the human health of Puerto Rican and African American women by understanding the physiological and molecular mechanisms that mediate the beneficial effects of moderate alcohol use. Specifically, we will investigate if moderate drinking affects the activity of the endocannabinoid system and the impact of endocannabinoid tone on bone loss. Preliminary studies from our laboratory demonstrate that ethanol modulates the activity of the endocannabinoid system and that this modulation is associated with changes in bone turnover markers, bone mineral density and strength. In order to gain further insights on the mechanisms that underlie this response we will determine the mechanisms of action of the most studied cannabinoid receptor, the CB1R and its possible role in the consequences of ethanol ingestion on bone. It is thought that ethanol activates the endocannabinoid system via CB1R, but there is an increasing evidence that another receptor, the GPR55, may mediate some of the ethanol effects on bone.
